Наиболее радикальной попыткой переработать проект L-410 стал вариант L-610G, оснащенный турбовинтовыми двигателями General Electric CT7-9D мощностью 1750 л. с. и рассчитанный на перевозку 40 пассажиров. Самолет был поднят в воздух 28 декабря 1988 года, однако после покупки "Let" компанией "Ayres" и последовавшей затем процедурой банкротства реализация проекта была прекращена. Удалось построить только семь самолетов данной модели.

Успех L-410 побудил LET разработать более крупный вариант L-610 - 40-местный региональный авиалайнер, схожий с предшественником, но имевший ряд важных усовершенствований. Продаж не было, и программу закрыли.

The fifth prototype Let L-610M 40-seat transport, 0005 (ex OK-136), has been delivered to Vyzkumny Ustav 030 (VU-030), the Czech Air Force test unit, based at Prague/Kbely airfield for trials. It is anticipated that some 8-12 L-610Ms will be bought by the air force to replace four An-24s, four
An-26s and some L-410s in the transport role.
